AM-694 (1-(5-fluoropentyl)-3-(2-iodobenzoyl)indole) is a drug which acts as a potent and selective agonist for the cannabinoid receptor CB1, with a Ki of 0.08nM at CB1 and 18x selectivity over the related CB2 receptor.[1] It is unclear what is responsible for this unusually high CB1 binding affinity, but it makes the 18F radiolabelled derivative of AM-694 useful for mapping the distribution of CB1 receptors in the body.[2]. No public data about AM-694 metabolism is known.

Heb geen ervaring met die specifieke cannabinoide, maar het effect zal wel in de buurt zitten van andere synthetische canna's, zoals de JHW series. Hier heb je iig een waarschuwing die op BL was gepost m.b.t. AM-694.

The metabolism of all indole cannabinoids involves loss of the indole N substituent, in the case of JWH 018 this is a pretty harmless alkyl chain. However in the case of AM-694 the N-substituent is a terminal fluoro alkyl, (omega fluoro pentyl) most terminal alkyl fluorides are exceptionally toxic as they metabolise to fluoroacetic acid.

w fluoroalcohols are also highly toxic 4-fluorobutanol has an LD50 in mice of 0.9mg/kg which makes it as toxic as some war gases.

whilst it is highly unlikely that sufficient fluoroacetic acid will be created by the metabolism of AM-694 to kill the consumer but fluoroacetate is dangerous even in sub lethal doses as it interferes with the generation of energy by cells. Fluoroacetate is used to kill vermin. In fact it completely screws up the Krebs cycle. AM-694 would effectively deliver this dose of fluoroacetate to the brain which is the most energy sensitive organ in the body. Repeated sub lethal exposures to fluoroacetate in animals have been linked to increased mortality liver damage, brain damage and damage to the reproductive organs. Fluoroacetate is regarded as a cumulative poison.

Until proper toxicological studies are available, it should be used as a research ligand for in- vitro use only.

Terminal N-fluoroalkyl chain acts differently from a normal alkyl chain found in JWH-018/-073, and this potentially causes the molecules to form covalent bond with the CB1-receptor (irreversible activation which leads to receptor internalization, and possible neurotoxic effects). N-fluoroalkyl group is metabolized into unbound molecule from AM-694, this makes it a potential alkylating agent which cause cancer. Also i'd like to test what AM-694 smoke/vapor contains before making anybody smoke it..

In a nutshell, AM-694 is quite radically different from JWH-018 and -073, and no human in vivo testing has been done other than a couple of user reports."
